Patient-derived xenografts, the cancer stem cell paradigm, and cancer pathobiology in the 21st century

The cancer stem cell (CSC) paradigm acknowledges the phenotypic and functional heterogeneity observed in solid tumors. This review discusses patient-derived xenografts, implications the CSC paradigm has for pathobiologists, and calls for greater collaboration between institutions, physicians and scientists to more rapidly advance our collective understanding of cancer.

Linking metabolism and epigenetic regulation in development of hepatocellular carcinoma

Hepatocellular carcinoma is driven by epigenetic changes that arise from exposure to environmental factors, rather than coding sequence mutations. Emerging studies reveal the underlying metabolic processes that are abrogated in cancer and lead to the altered flux and availability of key metabolites important for epigenetic processes. This review article discusses how altered liver metabolism affects long-term epigenetic processes which lead to the development and progression of HCC.

Resolvin D1 reduces deterioration of tight junction proteins by upregulating HO-1 in LPS-induced mice

Effective therapeutics are urgently needed for treatment of pulmonary edema. This paper demonstrates that resolvin D1 relieves pulmonary edema, restores pulmonary permeability and reduces disruption of tight junction proteins in lipopolysaccharide-induced acute lung injury of mice, at least in part, by up-regulation of heme oxygenase-1 expression.

Morphological and functional changes in a new animal model of Ménière’s disease

A new animal model yields insight into the mechanisms of water homeostasis in the inner ear and the vertiginous attacks of Ménière's disease. Animals receiving electrocauterization of the endolymphatic sac and desmopressin, a vasopressin type2 receptor (VP2R) agonist, show severe endolymphatic hydrops in the cochlea and the saccule, balance disorder, and spontaneous nystagmus. These abnormalities are similar to those in patients with Ménière's disease.

Urinary tract obstruction in the mouse: the kinetics of distal nephron injury

Fetal urinary tract obstruction impairs development, differentiation and maturation of the kidney. The authors use a postnatal mouse unilateral ureteric obstruction model, and show that distal nephron pathology is a significant component of unilateral ureteric obstruction. They demonstrate that this model replicates many of the key pathogenic features of fetal obstruction, making it a suitable and powerful model for investigating early medullary and distal nephron injury.

Protective role of tumor necrosis factor (TNF) receptors in chronic intestinal inflammation: TNFR1 ablation boosts systemic inflammatory response

Tumor necrosis factor (TNF) deficiency exacerbates the severity of experimental chronic colitis. TNF receptor 1 (TNFR1) −/−, but not TNFR2−/− mice, show increased mortality and augmented systemic inflammatory response compared to wild-type littermates. Granulocyte-macrophage colony-stimulating factor (GMCSF) is a key mediator in this process. Signaling via TNFR therefore plays a protective role in chronic intestinal inflammation, and the lack of TNFR1 augments a systemic inflammatory response in GMCSF-dependent manner.

A small interfering RNA targeting Lnk accelerates bone fracture healing with early neovascularization

Lnk is an intracellular adaptor protein expressed in hematopoietic cell lineages, and Lnk-deficient mice demonstrate enhanced hematopoietic reconstitution. A new role for this negative regulator of hematopoietic cytokine signaling is described in this study: Down-regulation of Lnk contributes to a favorable environment for healing bone fractures by enhancing vasculogenesis and osteogenesis.

Subtraction micro-computed tomography of angiogenesis and osteogenesis during bone repair using synchrotron radiation with a novel contrast agent

This paper describes vascular casting with a novel ZrO2 compound and subtraction micro-computed tomography, using two monochromatic synchrotron lights with energies above and below the k-edge of ZrO2. This technique is suitable for quantitative three-dimensional imaging of angiogenesis and bone regeneration during early-stage bone healing, demonstrating its potential for exploring angiogenesis-coupled bone healing process.
